What is Acalakast?

Acalakast is a new-generation Bruton’s Tyrosine Kinase (BTK) inhibitor, used in targeted therapy against white blood cancers, specifically B-cell malignancies.

Uses of Acalakast:

  • • Chronic Lymphocytic Leukemia
  • • Lymphocytic Lymphoma
  • • Mantle Cell Lymphoma

How does Acalakast work?

In B-cell cancers, abnormal B-cells grow and spread uncontrollably. Acalabrutinib blocks the kinase enzyme (BTK), which disrupts the signaling pathway responsible for B-cell division and growth.

Common Side Effects:

Headache, nausea, vomiting, constipation, diarrhea, joint pain, abdominal pain, and fatigue.

Serious Side Effects:

Fever, sore throat, blood in stool, infections, and fast/irregular heartbeat.

Warning:

Not to be used in patients with known hypersensitivity to Acalabrutinib or any of its excipients.

Precautions:

Use with caution in:

  • • Renal insufficient patients
  • • Breastfeeding mothers
  • • Pregnant women

Common toxicities include:

  • • Neutropenia – Increased risk of infections
  • • Thrombocytopenia – Risk of bleeding
  • • Anemia – Decreased red blood cell count

Other toxicities: Atrial fibrillation, bleeding, hypertension, skin infections

Indications for Use:
