What is Acalakast?
Acalakast is a new-generation Bruton’s Tyrosine Kinase (BTK) inhibitor, used in targeted therapy against white blood cancers, specifically B-cell malignancies.
Uses of Acalakast:
- • Chronic Lymphocytic Leukemia
- • Lymphocytic Lymphoma
- • Mantle Cell Lymphoma
How does Acalakast work?
In B-cell cancers, abnormal B-cells grow and spread uncontrollably. Acalabrutinib blocks the kinase enzyme (BTK), which disrupts the signaling pathway responsible for B-cell division and growth.
Common Side Effects:
Headache, nausea, vomiting, constipation, diarrhea, joint pain, abdominal pain, and fatigue.
Serious Side Effects:
Fever, sore throat, blood in stool, infections, and fast/irregular heartbeat.
Warning:
Not to be used in patients with known hypersensitivity to Acalabrutinib or any of its excipients.
Precautions:
Use with caution in:
- • Renal insufficient patients
- • Breastfeeding mothers
- • Pregnant women
Toxicities:
Common toxicities include:
- • Neutropenia – Increased risk of infections
- • Thrombocytopenia – Risk of bleeding
- • Anemia – Decreased red blood cell count
Other toxicities: Atrial fibrillation, bleeding, hypertension, skin infections
Indications for Use:
Acalakast is a prescription medication to be used under medical supervision only.
While generally well tolerated, it requires regular monitoring for blood counts, infections, bleeding risks, and cardiac toxicities.
